Abstract

Objective  To establish an ultra-performance liquid chromatography (UPLC)characteristic chromatogram and multi-component quantitative method for Fructus Viticis Negundo, to identify the differential components between it and the common adulterant Vitex Fructus, and to investigate the quality transfer rules from decoction pieces to standard decoction.

Methods  The characteristic chromatogram of Fructus Viticis Negundo was established by UPLC, the component differences between Fructus Viticis Negundo and Vitex Fructus from different sources were analyzed by chemometrics analysis. A method for the simultaneous determination of 9 components in Fructus Viticis Negundo was developed, and the transfer law of these major components from decoction pieces to standard decoction was investigated.

Results  The UPLC characteristic chromatogram was established, and 9 components were identified. Through cluster analysis and partial least squares discriminant analysis, the differential markers between Fructus Viticis Negundo and Vitex Fructus were identified as peak 2 (protocatechualdehyde), peak 5 (vanillic acid), peak 6, peak 10 (vitexin), peak 11 (isovitexin), peak 12, and peak 13. The average transfer rates of protocatechuic acid, protocatechualdehyde, p-hydroxybenzoic acid, vanillic acid, isoorientin, orientin, agnuside, vitexin, and isovitexin from the decoction pieces to the standard decoction were 74.48%, 79.43%, 53.95%, 65.12%, 22.78%, 43.22%, 33.90%, 39.50%, and 43.80%, respectively.

Conclusion  The UPLC characteristic chromatogram established in this study, combined with a multi-index content determination method, can effectively distinguish between Fructus Viticis Negundo and Vitex Fructus, and clarify the transfer law of the main components in the standard decoction, providing a scientific basis for the preparation process of Fructus Viticis Negundo formula granules.
